ABSTRACT

Objective To discuss the significance of the detection of interferon-γin the diagnosis of active joint tuberculosis. Methods 5 kinds of specific proteins of mycobacterium tuberculosis were used as stimulating protein, the chemiluminescence enzyme immunoassay was used to test the content of interferon-γ. The interferon-γof 35 cases of patients with active joint tuberculosis and 35 cases of healthy people were detected. The difference of the sensitivity, specificity, positive predictive value, negative predictive value, positive likelihood ra-tio and negative likelihood ratio of 5 kinds of proteins to the diagnostic of active joint tuberculosis were compared. Results CFP10/ESAT6 fusion protein, Rv2041c protein, Rv0057/1352 fusion proteins, Rv1419 protein and Rv1656 protein were used as stimulating protein, the sensitivities of active joint tuberculosis by the whole blood interferon-γassay were 77.1%, 68.6%, 71.4%, 74.3%and 65.7%. The specifici-ties were 65.7%, 54.3%, 57.1%, 60.0%and 62.9%. The positive predictive values were 62.9%, 60%, 62.5%, 65%and 63.9%. The negative predictive values were 74.2%, 63.3%, 66.7%, 70%and 64.7%. The positive likelihood ratios were 2.25, 1.5, 1.67, 1.86 and 1.77. The nega-tive likelihood ratios were 0.348, 0.579, 0.5, 0.429 and 0.545. Conclusion The detection of interferon-γhas certain significance to joint tu-berculosis. But it is not as an index of diagnosing joint tuberculosis.

ABSTRACT

@#Objective To evaluate the antibiotics release characteristic and osteogenesis ability of anti-tuberculosis composite graft by observing the healing effect of segmental radial defect in rabbits.Methods Rabbit decalcified bone matrix(DBM) was made under the method of Urist and was combined with rifampicin-gelatin solution. The antibiotics release characteristics were observed by PBS release test in vitro and K-B disc diffusion test in vivo. The osteogenesis ability was measured by critical radius bone defect of rabbit.Results It was detected in release test that the antibiotic releasing time could maintain at least 28 days in vivo and in vitro and the release ratio was 87%. The rabbit critical radius bone defect could be repaired by this anti-tuberculosis composite graft perfectly.Conclusion Good antibiotics release characteristics and bone formation ability showed during this anti-tuberculosis composite graft and it definitely could be used widely in further clinics.

ABSTRACT

Objective To investigate the gene expression changes in normal and degeneration lumbar intervertebral disc in humans, providing information for clinical. Methods The PCR products of 4096 human genes were spotted onto a kind of chemical-material-coated-glass slides. The total RNAs were isolated from the tissues. Both the mRNAs from the degeneration and normal lumbar intervertebral disc in humans were reversely transcribed to the cDNAs, which used as the hybridization probes with the incorporations of fluorescent dUTP. The mixed probes were then hybridized to the cDNA microarray. After high-stringent washing, the cDNA microarray was scanned for the fluorescent signals and analyzed with computer image analysis. Results Among the 4096 targets,there were 706 genes whose expression levels differed between the degeneration and normal lumbar intervertebral disc in all cases,comprising 298 up-regulated and 358 down-regulated ones. Conclusion DNA microarray technology is an effective technique in screening for differently expressed genes between the degeneration and normal lumbar intervertebral disc. Cell apoptosis plays an important role in the process of lumbar intervertebral disc degeneration.
